Stained CD8 from culture

Dear Folw-ers,

I stained murine splenocytes, from culture, with CD4 and CD8 antibodies. I
can't see a "compacted blot" on CD8 positive population, I see several dots
but not compacted. On CD4, everything is normal. CD8 stained cells from
murine freshly isolated splenocytes present "compacted blot" like CD4.
But, in culture, this phenomenon not occurs like on freshly isolated cells
(on CD4 this "compacted blot" occurs). Anyone have an idea what is happened
?? I know that CD8 population is lower than CD4, but is it the problem ?? I
use the same antibody concentration for staining CD4 and CD8. Both
antibodies are obtained form Pharmingen.
Sincerely

Victor Brasil
