Ernst,

human ficoll isolated PBMC's also contain monocytes. Assuming you
plan not to gate on those - ie. you're only looking at lymphocytes -
you can expect ~65 +/-5% CD3+ cells (healthy controls of course).

in my experience ficolling tends to reduce number of NK cells
fraction but CD3+ cell counts seem unaffected.

finally if you are gating on everything - IE. all your PBMC's, you
can expect great variation in CD3 %tages.
