Has anyone out there evaluated the Coulter Flow-Count bead and TetraCHROME reagents vs. CBC/diff and TetraCHROME using heparinized whole blood? We recently did a parallel study and found the single platform Flow-Count absolute number of CD4, CD8, and CD3 cells to be consistently 10-20% lower that the standard dual platform calculated absolute numbers. The correlation coefficient (.96 for CD4) doesn't look bad, but the negative bias is troubling. Coulter tech. service suggests that is may be due to the anticoagulant heparin. Their own studies were done using EDTA (not an option for us). I'd appreciate the benefit of your experience and insights.
