Pharmingen DimerX

Greetings to all on the list

Anyone had a go with Pharmingen's DimerX product yet? It's
recombinant soluble dimeric mouse class I molecules fused with mouse
Ig that can be loaded with peptide to detect antigen-specific T cells
ala MHC tetramers.

I'd like to hear from anyone who's used it with mouse spleen and
lymph node preps to detect antigen-specific CD8 cells. The class I
dimers are picked up with an anti-mouse Ig conjugate--looks like it
works great with T cell clones but what about when B cells, macs and
DCs (lots of surface Ig and Fc receptors) are in the prep? Can all
that background binding be successfully blocked with confidence to
leave only the peptide/MHC-binding cells?

Thanks

Matt
