Dear Flow-er, I am going to evaluate and perform the TdT staining on the acute leukaemia samples. Having retrieved some of the old mails and journals, I found that performing TdT staining in Flow cytometry is not quite easy. Could anyone so kind to share some of your experience on the following: 1. Which kind of clones is better, monoclonal or polyclonal, FITC or PE conjugated, brand? 2. Does the BC Intraprep suitable for the permemablization and fixation step? Any other suggestions? 3. What other significant consideration on performing this staining should be concerned? I know that it takes times to test my protocol but your valuable suggestions will certainly help. Thanks a lot. WK LEE UCH, Hong Kong
