Richard Haugland and Howard Shapiro state correctly that there is no fluorochrome that specifically stains RNA. The specific staining of RNA, however, can be achieved immunocytochemically. Incubation of cells with BrU labels specifally only RNA. Some antibodies against BrdU crossreact with BrU - the one that does it is provided by Phoenix Flow System. This method, first described by Jensen et al., (Cytometry, 14;455-58,1993; see recent protocol by these authors in Current Protocols in Cytometry) provides indeed very specific RNA labeling. I am attaching our recent paper demonstrating the use of this method to follow RNA segregation during apoptosis Zbigniew Darzynkiewicz Brander Cancer Research Institute New York Medical College 19 Bradhurst Ave. Hawthorne, NY 10532 tel: 914-347-2801 fax: 914-347-2804 http://www.geocities.com/z_darzynkiewicz <<RNADNA.pdf>>
