Dear Lynn,

Beside tetramers, there are other new approaches to detect antigen specific
T cells based on cytokine production (i.e. cytokine release assay from
Miltenyi Biotec) which works also with a frequency = or <0,1% positive
cells. If you can achieve a very good negative control with low background
(<0,01%) then the assay is pretty reliable.
