I have a question about FACS for DNA content in Candida albicans. Is it possible that a candida hyphal cell, which consists of a long string of cells (in my case 2 cells attached) separated by septum, with individual nuclei in each compartment are counted by the detectors as 2 cells with 2n content each rather than one cell with 4n content. the nuclei are about 20-30 micron apart, but, as I mentioned, are attached on one long hypha.
