On a BD Calibur or FACScan, the endreduplicated cells can usually be
clearly distinguished on a dotplot of  width versus DNA area. 

On the Beckman Coulter XL or Elite, a plot of DNA peak versus DNA area will
show these cells. It is often easier on these machines because the laser
beam has a narrower width.

I have attached a couple of examples taken from one of the lectures on my
Internet course. The region encompasssing single cells ahs nee extended to
include a small percentage of endoreduplicated cells.

I have seen endoreduplicated cells quite commonly in cell lines. It can be
quite a problem as often the properties of the cells change and the worker
does not realise that his/her long term cultures are changing.

I always advise people to check the ploidy of long term cultures regularly;
but they seldom take my advice!
