All, I am posting this message for a friend of mine who is involved in immunohistochemistry. She is trying to stain mouse B lymphocytes in tissue that has been formalin fixed for over 10 days. She has been using a CD79 antibody from two different sources and has any luck. She has been able to stain T lymphs in the same tissue, and the control tissue, canine spleen has worked well with the CD79 as well as the Tcell antibody. She is thinking that the B antigen sites have been denatured by the very long formalin fixation time. Any thoughts, suggestions, etc.? Thanks in advance.
