Dear Flowers I have recently started doing cell cycle analysis on the budding yeast, Saccharomyces cerevisae. I have used the protocol of Fortuna et. al. in Current Protocols in Cytometry. This method uses SYBR green I and produces a very nice cell cycle distribution. However, interpreting it is somewhat different then mammalian cells which I am used to dealing with. The cells exhibit budding (up to two per mother cell). I have attached a picture of my histogram and was hoping someone experienced in yeast cell cycle could comment/identify these populations.
