Dear flowers, I am trying to measure intracellular TNF-a production of bone marrow-derived macrophages in response to LPS stimulation. Could someone please make comments on two questions: 1) for this kind of experiment, what's the best way to get macrophages off the culture dish: rubber policeman, ice-cold PBS plus EDTA, trypsin, etc.? 2) what's the optimal time point for measurement?
