Distinguishing cell populations

I was wondering if anyone could help with this problem.  We have just
started flow cytometric analysis in our lab and are looking for different
methods to distinguish between different cell populations in FACScan.  Does
anyone know of a method that is a more objective way of distinguishing
between cell populations other than be eye?
