I am trying to use FACs analysis on rat aortic smooth muscle cells. Unfortunately the cells do not detach with EDTA/no Mg,Ca conditions, and if I force them off with a high velocity stream of media, many of the cells are damaged. I am trying to detect a cell surface proteoglycan, so I do not want to use trypsin to detach the cells. Does anyone have any ideas as to what I can use to get the cells in suspension?
