need advice on choice of 2nd step antibodies

Jackson Immunoresearch has an abundance of secondary Abs that are F(ab')2
fragments that don't bind to Fc receptors AND have also been adsorbed
against multiple species.  I have had excellent results whenever I have
used their antibodies.  It sound like you have a real problem with your
current secondary Ab and I strongly suggest you try one of Jacksons.

The other company that has some isotype specific secondary Abs is Southern
Biotechnology Associates, Inc. , phone: (205) 945-1774.  We just bought a
goat anti-mouse IgG1-HRP conjugate for Westerns and it was great.

You can try any fluorescent combo you want.  Your problem is your
background staining with the FITC-conjugated secondary.  Secondary only
should never be so high.  Try out the above companies.  I am certain it
will improve your results.  Good luck.
