In cell cultures, cells not infrequently endoreduplicate. There will then
be a tetraploid cell cycle imposed on the diploid cell cycle. You can most
clearly demonstrate endoredupliation by staining for cyclin B1.

I have assumed that you have eliminated cell clumps from the analysis,
these are the most trivial explanation for a DNA content >G2.
