FGFR staining

Dear list,

I am interested in looking at the expression of fibroblast growth factor
receptors (transmembrane) in bone cells.

I have primary antibodies to the 4 FGFRs that recognise the C terminal tail
(cytosolic).  Therefore, I need to permeablise the cell membrane to allow
the antibody to bind.

Does anyone have a protocol suitable for FGFRs?

Also, most protocols call for 1 million cells to be divided into 50
microlitre aliquots for fixing and staining.  Is this a sufficient number of
cells per 50 microlitre aliquot or should I use more?
