Just a general question: While charts of fluorochrome spectra are easy enough to come by, could somebody point me in the direction of a table of (relastive) fluorochrome intensities? Quantum yield is a clue, but doesn't always reflect the real world when put to practice. For example, PE is brighter than FITC. What of PE-Cy5 vs PerCP, APC etc. Would a Cy5 excited at 633 be brighter than a a PE-Cy5 tandem excited at 488 or vice versa? If using Cy7 (or conjugates thereof) are the (standard) PMTs of BD, Coulter, and Cytomation instruments sufficiently sensitve in the far red, or should the PMTs be upgraded. In other words, in selecting fluorochromes for dim (surface) markers, what would be the order of selection? And whatever info you care to share.... Many thanks, Claude
