Hi, A very basic question to the group. Most probably due to my inexperience I am unable to pin down a ref other than to Hoechst 33342 (Ho). Basically I want to sort live cells into cell cycle subsets (G1 and anything but) and them examine various transfection protocols on on the sorted populations. For such, staining on the basis of DNA content appears to be appropriate then I would flow sort the transfected populations in a couple of days or so for eGFP using flow again. Is a Ho based protocol the only option or are there textbooks of alternatives on the stuff.? Does Ho affect further 'normal' cell cycling characteristics in such a situation?
