Dear flowers, one of our investigators would like to perform flow staining on human sperm. The purpose is to identify autoantibodies in various human fluids. We already performed sperm viability using PI, but staining with human fluids followed by a rabbit anti-IgG or anti-IgA-FITC gave us very unconsistent results (when compared to classical beads aggregation assay). Is it possible to detect antigen present on the head as well as on the flagellum of the cell? Any experiences, protocols or references will be welcome. Thanks in advance.
