Greetings, I am writing on behalf of an investigator who wants to assess RNA in different populations of mouse hepatocytes using a quantitative DNA stain. The latter must not affect the RNA so Hoechst 33342 seemed the obvious choice. After numerous permutations of the Hoechst staining protocol (varied concentrations, staining times and temps, addition of Verapamil ) 2N, 4N and 8N populations could not be discriminated. If anyone has successfully stained these cells using Hoechst, or if someone can suggest a suitable alternative, we'd appreciate your comments.
