Compensation of Dendritic Cells

Dear Madams and Sirs,

I have problems with autofluorescence of my murine bone morrow-
derived dendritic cells. I received now the FACS settings for a
Beckton-Dickinson FACScan which should deal with the
autofluorescence of the DCs. The settings are as following:

FSC Voltage: E00 1.00	Linear
SSC Voltage: 303 1.83	Linear
FL1 Voltage: 457 1.00	Log
FL2 Voltage: 519 1.00	Log
FL3 Voltage: 732 1.00	Log

Compensation:
FL1 -	0.1% FL2
FL2 - 84.0% FL1
FL2 -	1.0% FL3
FL3 - 20.8% FL2

I would now like to know whether a compensation of FL2 - 84.0%
FL1 is acceptable because I was told that the compensation should
not be over 50%.
Are there other ways to overcome the problem of autofluorescence?

Best regards,

Karen Wahl
