As they say in real estate: Titration, titration, titration. The biggest variable will be to optimize S/N by titrating your serum. Depending on the species of your cells, serum and secondary Ab (as well as if it is adsorbed for other species besides the serum donor species) you may wish to try blocking with IgG (from a different species) at high (>> mg/ml) concentrations. Five % non fat dry milk may also be worth trying. > ---------- > From: Isabell Schmitt > Sent: Thursday, May 11, 2000 11:32 AM > To: Cytometry Mailing List > Subject: serum-staining > > > Dear Flowers, > > does anyone have experience in staining cells with serum instead of > unconjugated first > antibody for FACS analysis or MACS-sorting?
