If I remember my basic microscopy lectures; live cells exclude the dye by virtue of an intact cell membrane, non-viable in this case means that membrane channels are passive and incapable of preventing dye entry, or that membrane structure has broken down. If you want further measures of cell viablilit you can use LDH enzyme release ( a measure of total membrane integrity loss), and the MTT dye assay ( a measure of metabolic activity) Craig Turner IBGRL ---------- From: peter.kierulf@ioks.uio.no To: cyto-inbox Subject: Reference- TRYPANE BLUE EXCLUSION Date: Wednesday, March 29, 00 23:30 <<File Attachment: ALTERNATIVE.RTX>> Hi there!, We as many others have been using trypane blue staining to account for number og dead cells. We have tried in vain to find a basic reference that gives information as to why the cells become trypane blue positive when they are dead. Can anyone help out?? Thanks Peter Kierulf,MD PhD.
