Dear flow experts, One of our staff is interested in measuring DNA content in Dictyostelium discodium amoebae using flow techniques. The problem is Dictyostelium also carry fairly good amount of mitochondrial DNA , around 30% of its total DNA . It is obvious, therefore measuring total DNA does not relate to cell cycle alone . Can anyone suggest a solution to exclude the contribution from mitochondrial DNA from the total, before or after DNA measurement?. Thanks in Advance P.S.Manogaran.
