Other example about "quality" of some reagents. I wanted to label rat CD4 +
other two markers (3-colors). I made a mistake buying the isotype controls: they
were supossed to be used only with human. In fact one of the isotype controls
was an anti rat-CD4 (exactly the same clone that the "positive" anti-rat CD4 as
it was described in the data-sheet). In this point I was thinking that I have
two bottles of the same reagent: wrong!! The anti-rat CD4 used as human isotype
control was not staining rat CD4. After this experience I think that isotype
controls can be a risky bussines: how do you know what do you have inside the
bottle when you can not check what is staining the isotype control?

FLOWERS:

To begin this is not a slam against antibody manufacturers or suppliers.

We have been experiencing an ongoing problem. We have purchased goat anti
Human IgG (conjugated to three different fluorochromes one red and two
green, and unconjugated) We have even labeled the unconjugated with alexa
488.

Here is the problem none of these reagents is specific for human IgG. We
have extensively tested this. We have talked with two of the suppliers at
length.

My question: Is anybody seeing similar product failures. It would appear
that the suppliers of the antibodies (secondary reagents as well as others)
do not manufacture the clones. This makes sense when you think about the
number of antibodies any given supplier markets. My thought is that most of
the antibodies are being produced by say one manufacturer and the antibodies
are sold to the various suppliers, who then conjugate or don't and sell to
the users.

>From what I see this current lot of goat anti Human IgG is not any good and
it was supplied to the suppliers
that I have tried.

I bring this to your attention, because I would bet most of you use the same
three suppliers as me. One in Seattle, one in the South and one in Maryland.

Just a little FYI. If any want to discuss this further, please contact me by
Email
