Hi Maryalice, I used to put the tissue in tripsin-edta for 30-60
minutes at
37 C, and after that is easy to separate epidermis from dermis. We
get good
recovering of Dendritic cells. You can also use dispase or DNase in order
to get the full cell suspensions.
