Dear colleagues, We are trying to do ploidy analysis on various hematoprogenitor cells from mouse spleen and bone marrow. Every time we've tried to react our cells with FITC-mabs / fix and then add propidium iodide (for ploidy), we notice what looks like significant quenching of the FITC signal so that defining our phenotypes (for gating our ploidys) is next to impossible. Basically we are stumped at what to try next. Please let me know of a reliable procedure that might allow us to succeed doing this type of analysis. Thanks, Phil Marder
