intracellular markers of eosinophils

Last week Dr. Dawn Cooper inquired about markers of eosinophils. 
We have noticed that fluorescein isothiocyanate (FITC) is quite a specific
marker of eosinophils (Bedner et al., CYTOMETRY, 36: 77-82, see also the
photograph on the cover of the last issue - Jan 2000, of CYTOMTERY). At
2-200 nM concentration FITC stains eosinophils' granules very stronly while
concentrations > 500 nM are needed to stain other granulocytes, monocytes or
lymphocytes. Highly charged cationic proteins, components of the eosinophils
granules, such as eosinophil cationic protein (ECP) and major basic protein
(MBP) are most likely responsible for the high affinity binding of FITC
