A collaborator of mine has performed an in vivo depletion of CD8 cells in mice using clone 2.43. When he went to look for CD8 cells using PharMingen's 53-6.7 mAb they appeared depleted. Unfortunately, when we tried a simple blocking experiment in vitro, pre-incubation with 2.43 blocked subsequent 53-6.7 binding, suggesting that the "depletion" may simply be due to antibody cross-competition (the 2 mAbs recognize the same or closely related epitopes). Does anyone in the group know of other commercially available CD8alpha antibodies that do not cross compete with either 2.43 or 53-6.7? We cannot use CD8beta antibodies. Is there a mouse CD data base as exists for human CD antibodies?
