BD antibodies

We have noticed the same thing with the "TRUCOUNT" reagents. We have noticed 
that when you look at the CD45 percp and SSC. Most of the cells are falling 
in the first decade, in another word they are negative for CD45. The CD34 PE 
is also very weak  compared when you use 20ul or even 5ul of CD34 percp from 
a different bottle. I am not sure if they are diluting the CD34 so much as 
to keep the back ground staining very low, since it is suppose to be a Lyse 
no wash technique.
When i add 5 ul of CD45 and CD34 in addition to the recomended volume of 
TRUCOUNT reagents the staining looks very nice and bright and your CD34 
population stands out away from the rest of the cells. I think if they 
increased their concentration of the antibodies people will be able to use 
this reagents with crude bone marrow as well.  For this reason we stopped 
evaluating the TRUCOUNT reagnets, specially when we are paying four times 
the list price. I am not sure if somebody else had also a problem with the 
APC Calibrite beads.. there is one particular lot number that keeps failing 
the FACSComp on our FACSCalibur, and so far I have not heard any response 
from BD in Belgium for an explanation.
