Dear all, I am working with the Balb/c3T3 murine fibroblast cell line in order to detect apoptosis induced by metal compounds. I used an annexin V/PI kit from Immunotech. With this assay I sometimes got positive results, however, with many difficulties, uncertainties and without a good reproducibility. It seems that this annexin method doesn't work very well with my cell line. Because I am a beginner in this research field, I would like to ask you some suggestions regarding my problem. I also know that caspase-3 assay may be an alternative solution. By checking a suitable kit, I have found many methods such as fluorimetric or colorimetric assays as well as by flow cytometry. What is the difference between them? And which one could be best suitable for the Balb/3T3?
