Dear listers,
I had a couple of request for a summary of the responses received on this
topic so I decided to post it to the list.

Here is the summary but first an apology: the correct name for the source I
was trying to locate was Statens Serum Institute (Copenhagen) and not
Statens Institute. However, I phoned and e-mailed them, but still no answer
yet.

1. We use TET toxoid from Wyeth (lot# 112191 NO PRESERVATIVES!) with great
success (PBMC, 1-4 microg/mL, activation by flow day 3-5, 3HTdR uptake on
day 5). My attempt to get hold of the same molecule here in the UK was
unsuccessful.

2. We use Tetanus toxin, C-fragment, Calbiochem cat.582235
(www.calbiochem.com). Calbiochem also have tetanus toxoid and tetanus toxin.

3. Try this address: <mailto:folkehelsa@folkehelsa.no> . Is the Norwegian
National Institute of Health.

4. We have used purified tetanus toxoid that we obtained from the
Massachusetts Public Health Biologic Laboratory (USA), web site:
http://www.umassmed.edu/service/massbiolabs/ phone: 617-983-6400 Fax:
617-983-6445

as of 1997 :
column purified tetanus toxoid
lot # CP-55
1 mg 150 US$
5 mg 675 US$
10 mg 1350 US$

This has worked quite well as a recall antigen. The work we have done was
in nonhuman primates. We have found that if they are immunized with alum
precipitated tetanus toxoid the recall as measured by proliferation is an
adequate form of measurement. If they are immunized with purified tetanus
toxoid without an alum adjuvant, you probably won't be able to detect any
response via proliferation. If you run into this problem I can forward you
a protocol to measure precursor frequency on the flow cytometer.

5. I think List Biological Laboratory in Campell, CA may have what you want.
If not, they should be able to tell you where to go. (http://listlabs.com)
Indeed they have it in 25 and 100 UG sizes.

6. Though I have not used tetanus toxoid from this source, Toxin Technology,
Inc. has been a good source for us for other uncommon antibodies and such
for flow analysis. They are located in Sarasota, FL, USA-- email
toxtech@att.net, phone
941-925-2032, fax 941-925-2132.

7. We use Absorbed Tetanus Vaccine BP (Pasteur Merieux from MSD) 5 x 0.5ml
ampoules per pack. We buy this direct from our local chemist. We tend to use
it at 1:1000 to 1:2000 with 2xE5 cells (PBMC's post lymphoprep) per well and
get between 5000 & 20,000 counts with a 6hr pulse at Day 5.

8. We use tet.tox from CSL in Melbourne Australia, please try their web
page, it is working.

9. This is a difficult reagent to get hold of-Connaught Pasteur/Merrieux
used to have it available with a thimerisol preservative that you would
probably need to dialyse. I don't have the contact information any longer,
but it shouldn't be too hard to find. I believe they make the major tetanus
vaccine used in the US, and are located in Pennsylvania.

10. I also have found one more source: CN biosciences cat. n. 582231 (25UG).

Happy 2002 again,
Mara
