Intracellular staining reagents question (BD)

Hi all,

I did cytokine staining with activation and skipped the lyse step,
adding perm. solution(BD) directly after surface staining. As I
expected - it worked, RBC's were lysed and I got a very clear scatter
picture even without gating on CD45 bright cells.

Does anyone else do their cytokine assays in whole blood this way?
Are there any pros/cons ?

I would especially like feedback from any of the BD scientists since
they probably have a lot more experience with these reagents.

regards,

Maciej
