We also do CD34 isolation from fresh and frozen cord blood and mobilized peripheral blood routinely. The MACS kit uses QBend 10 Ab which is against a class 2 epitope. HPCA-2 (BD clone) or the Ab from Pharmingen are against a class 3 epitope which is the most stable and can be used to stain MACS isolated cells. Since the beads are very small they do not affect the scatter properties of the cells, but sometimes they appear as debris and need to be gated out.
