Dear Flowers, I have been trying to isolate cells from bovine placenta (cotyledon specifically) and examine cell surface markers using flow cytometry. I have several (many ?) problems. One problem is keeping samples. I cannot get placental samples easily, then I would like to keep placental tissue for later use if possible. Now I use only "fresh (taken just after expulsion or peeled from uterus after calving)" or one day old samples kept at 4 C. Does anyone know how to keep placental tissue for a long time so that I can isolate cells later and stain cell surface markers and intracellular cytokines? If tissue storage is impossible, how about isolated cells? Can I keep these cells for a long time for later staining?
