I have an idea. What if we were to include some basic information about our analyzer/sorter with any questions we posed to the list? I learn so much from the list, but I think we might get better responses to our questions, more quickly, if we included pertinent information. For example, if I ask a REALLY broad question (and we DO see these!): How do I do cell cycle analysis by flow? ...people at least have a jumping-off point to answer the question if I include some basic info: 1. I'm using a standard FACScan. 2. I'm using a FACSVantage with an Enterprise 621 laser. 3. I'm using a Coulter Elite with a Cyonics Argon laser (488 only) and a M-G 30 mW HeNe. 4. I'm using a MoFlo with Enterprise 621 for 488 & MLUV, plus an I-70 Argon/Krypton (647). Each of these additional pieces of information could generate much more specific responses. Additionally, I guess that more people might respond if they feel that they don't have to cover ALL the bases! I was recently educated (again!) by the responses to the GFP/YFP question. I have pretty good flexibility in terms of laser line selection, so I didn't immediately think to change filters for the GFP/YFP problem. I started thinking of ideal excitation wavelengths to better excite the two proteins independently. So much to learn, so little time! Thanks for listening!
