Dear flowers, I have two years ago published a paper about that stuff with the title Fluorescence enhancement of DNA-bound TO-PRO-3 by incorporation of Brdu to monitor cell cycle kinetics. Cytometry 37:221-229 (1999). It explains how TO-PRO-3 can be used in a similar way as the Hoechst quenching technique, however, an expensive UV laser is not necessary. In principle it works with a FACSCALIBUR, bit I have not extensively tested it.
