Dear Flowers: Has anybody done Calpain analysis by flow? Are there reagents for such a test that I should consider? I have found that Molecular Probes does a Aminochloromethylcoumarin (CMAC) peptidase substrate called t-BOC-Leu-Met-CMAC (cat no. A-6520), that supposedly calpain is able to cleave to produce a fluorescent product. Are there any recommendations or abvice on the efficacy of this product, particularly regarding specificity with repsect to calpain. Scott Tighe Vermont Cancer Center Flow Cytometry Core
