Dear Flowers, Does anyone have any experience with the Z1? I am using the Coulter Z1 to count human PBMCs at low concentration in various media. I set the volume gates to count particles between 35 and 160 fl, as recommended by the manufacturer, for PBMCs. However, this setting gives me a large number of counts above 160 fl, appearing to indicate substantial clumping, even though by eye, it looks like a nice opalescent suspension, and the medium contains 2mM EDTA, as recommended by Flowers to minimize clumping. Should I set the upper volume gate to a higher value? or are these two sizes, 35 and 160 fl, completely correct?
