Sathi,
The 4-color XL uses 530, 575, 610, and 675nm bandpass filters. APC excites at 633 nm
(HeNe), and emits at ~675 nm . . . the XL won't do it. The 4 color setup typically
used on an XL would be:
FL1 - - some form of fluorescein
FL2 - - some form of phycoerythrin (PE)
FL3 - - a PE-Texas Red tandem (Red613, ECD)
FL4 - - a PE-Cy5 tandem (Cychrome, TriColor, Red670).

I like to do a 4 color-analysis on Coulter-Epics/XL for lymphocytes. For
one of the marker, I have an APC-conjugates antibody. Could any one tell
me the emission/excitations spectrum for APC-fluorochrome and which channel
should I use for APC on Coulter Epics/XL cytometer. I will be using FITC
and PE for other channels and the info would help me to choose the fourth
color (ECD or PC-5).

I would appreciate any helpful comments.

Thanks
Sathi
