Flourescent labelled intracellular peptide

Dear Flowers:
There is a PI here that is trying to detect flourescence of a FITC labelled
peptide that has
been injected into live models.  They are certain the peptide has entered
the cells by the
positive response to treatment, however it is not being detected by flow.
It is, however,
seen under a flourescent scope.  They are seeing positive FITC when the
assay is
performed on cultured cells.  They have said that this has worked for them
in the past and
are curious as to ideas on detection of intracellular flourescence using a
FACSCalibur.  If
anyone can help it would be greatly appreciated.  You can respond to me, I
will relay the
information.
