curious finding during IL-10 experiment

I wanted to do a "test" run on new lot of antibodies we're going to
use for an intracellular il-10 measurment in monos (see previous
posts).

When it was all done, I found that CD33 was nowhere to be seen in
tubes activated with PMA (4 hrs). CD14 was there just fine. I was
expecting it to be the other way around.. I didn't use LPS b/c I know
that will block/shed my CD14's. With other activators I used today I
had normal results, so only the PMA raised an eyebrow. This is the
first time I used PMA to check cytokine production.

my PMA came from leftover bursttest kits by orpegen, I used a modest
.02mM final conc.

I wonder if anyone had similar findings or any words of wisdom?

Maciej
