Work in my department involves determining the intracellular cytokine stores of murine CD4 and CD8 cells. Following staining and assessment via flow, results to date have not demonstrated many positive cells, even in cell lines known to be producers of the two cytokines we are assaying for, IL-4 and IFN-gamma. Would anyone have any trouble shooting tips or know of any cell lines that are producers of these cytokines that come pre-stained for intracellular cytokines that we can use as a positive control.
