Dear Flowers, For sample preparation of either marrow or peripheral blood, how can I decrease the background staining. I have come across an AML case with abnormal population which is sticky and take up the isotypic control heavily. Even for the negative marker, the cells stained heavily and so the cut-off of pos/neg population depends greatly on the isotype control. Can washing step or incubation with 30% albumin helps?
