Reasonable CD34 and CFC correlations can be achieved between the pre cryo and post thaw counts on PBSC products. CD34 analysis must be performed using a no antibody wash method for the thaw sample at least (best for both) and a single platform "bead" method is worth considering. We use the Stem Cell Technologies, Methocult H4434 for our CFC assay, this has produced more reproducible results. I have included an abstract and some grafts.
