Jurkat cells and intracellular staining of IL4

 Dear Flowers,
 Hello and best wishes.
 We want to develop an assay for the detection of IL4 intracellularly in an
appropriate human T cell line (Th2 profile of cytokine expressing cell line).
At present we are using a Jurkat cell line and are not able to detect IL4
intracellularly even though PBMC show it.
 There are some doubts that I have and would be grateful for any help
regarding them.
  1. There are a couple of Jurkat derived cell lines but most of them are
stated to produce IL2 ( as mentioned in ATCC catalogue), has any one used
these cell lines for detection of IL4 after stimulation with Ionomycin and
PMA?
  2. What are the cell surface markers that define these cells?
  3. Are there any other human T cell lines that produce IL4 upon stimulation
with Ca2+ ionophores and PMA?
