People have looked at isolated mitochondria with rhodamine 123 and probably
other dyes such as JC-1; these, however, generally provide a relatively
crude estimate rather than a calibrated value of membrane potential.  A
more accurate measurement could probably be obtained using the ratiometric
technique with DiOC2(3) developed in my lab for bacteria by Dave Novo
(Cytometry 35:55-63, 1999); this probably won't work for mitochondria in
situ but should for isolated mitochondria.
