Paul, We've worked pretty extensively with cynos and have used both FACS Lysing Solution and the various Prepstations and reagents from Coulter (Q-Prep, TQ-Prep) with good results from both. We've also done gradient separations on cyno blood with fairly good separation, although in our hands, the procedure is much more time consuming (need to spin longer than human cells), the preps aren't always that clean (RBCs and granulocytes mixed in with the mononuclear cells) and I'm not convinced that the preps are any better than lysed samples. As for antibodies, check with Pharmingen and Coulter for cross-reactive clones. Good luck, Patrick B.
