Antibody Binding Capacity - Autofluorescence

Hi all,

I am trying to calculate ABC for cells with very high levels of autofluore-
scence.  Two questions:

1) Is it really accurate to simply subtract off the ABC for unstained cells?

2)  Is it OK to extrapolate outside of the calibration plot for ABC vs. 
channel number?
